In vitro Production of Plantlets from Node Culture of Oriental Melon and Muskmelon (Cucumis melo)






Abstract
The experiments were conducted to obtain basic information on propagation of seedlings of melons by in vitro node culture. Shooting from node and subsequent growth of shoots were well performed in MS medium with 0.2 - 0.5 ppm BAP. Shooting was retarded by addition of NAA but callus formation was promoted. Shooting and growth of shoots were promoted in the medium supplemented with below 1.0 ppm of BAP and Kinetin combination but culture efficiency in 2iP containing medium was varied to its concentrations. When different parts of the plantlet obtained by initial node culture were subcultured, nodes in upper part of the stem shooted very well, and the shooting was gradually decreased to lower part. Shooting was better in muskmelon than in oriental or weedy melons. Plantlets obtained by in vitro cutting rooted well in 0.05 ppm NAA and Gelrite containing medium. The seedlings grew well when transplanted in the pot after 2 weeks hardening, bloomed and set fruits.
